It was passover - when many people made a pilgrimage to what is now the Dome of the Rock, but at the time was The Second Temple - Jesus and disciples arrived to find it full of livestock and cattle traders and Money Changers (converting Roman denarri and Greek drachma into shekels).

It's significant because a) it represents the power of religion over commerce, and b) because it fulfills a prophesy earlier in the Bible

Selling stuff in a church or something? I don't remember, but he didn't supposedly deal with it in a very pacifist manner.
